ABSTRACT

OBJECTIVE: To establish the method for simultaneous determination of curdione, curcumenol, germacrone, furanodiene and β-elemene in Curcuma wenyujin, and to optimize its processing method in production place. METHODS: HPLC method was adopted. The determination was performed on ODS SP C18 column with mobile phase consisted of acetonitrile-0.05% phosphoric acid solution (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 214 nm, and column temperature was 30 ℃. The sample size was 10 μL. 18 batches of sample medicinal materials were prepared by steaming-drying (10, 20, 30, 40 min), or cooking-drying (drying temperature 50, 60, 70 ℃), and then the contents of 5 components were determined. Relative relational degree (ri) obtained by grey relational analysis method as indexes was used to optimize the processing methods of C. wenyujin in production place using. RESULTS: The linear range of curdione, curcumenol, germacrone, furanodiene and β-elemene were 25.56-409.0, 2.05-32.8, 6.36-102.0, 9.14-146.0, 8.62-138.0 μg/mL (r≥0.999 2), respectively. RSDs of precision, stability and reproducibility tests were all lower than 2.0% (n=6). Quantitative limits were 0.34, 0.62, 0.11, 0.14 and 1.20 μg/mL, and detection limits were 0.10, 0.19, 0.03, 0.04 and 0.42 μg/mL, respectively. The average recoveries were 101.3%, 98.7%, 99.0%, 99.6%, 96.4%, respectively (RSD<2.8%, n=6). The results of grey relational analysis showed that the relative relational degree (ri) from the processing technology of “boil for 10 min-drying at 50 ℃” was greatest, which had relatively small comprehensive effect on the contents of curdione, curcumenol, germacrone, furanodiene and β-elemene respectively. CONCLUSIONS: Established HPLC method can be used for content determination of 5 components in C. wenyujin. The results of optimization can provide reference for the selection of processing method of C. wenyujin in production place.

ABSTRACT

Objective To investigate the correlation between the infarction location and progressive motor deficits (PMD) occurrence.Methods The patients with middle cerebral artery(MCA) infarction within 24 h of onset without thrombolytic therapy were included.The National Institutes of Health Stroke Scale(NIHSS) motor item score increase ≥2 points of the base line within 7 d after stroke onset served as the PMD diagnostic criteria.The differences in clinical and laboratory data,and infarction location were compared between the PMD group and non-PMD group.The multivariate Logistic regression analysis predicted the risk factors of PMD occurrence.Results A total 121 patients with MCA acute cerebral infarction were included in the study and divided into the PMD group (45 cases) and non-PMD group (76 cases).The internal watershed infarction occurrencerate in the PMD group was higher than that in the non-PMD group (26.7 % vs.5.3%,p=0.001).The occurrence rate of penetrating arterial infarction (PAI) had no statistical difference between the PMD group and non-PMD group(42.2% vs.35.5%,P=0.463).PAI was further divided into perforating branch atheromatous disease (BAD) and lipohyalinitic degeneration (LD).The occurrence rate of BAD in the PMD group was significantly higher than that in the non-PMD group (28.9% vs.9.2%,P=0.005).The stepwise Logistic regression analysis indicated that watershed infarction [odds ratio (OR):9.750,95 % confidence interval(CI):2.828-33.612,P=0.000] and BAD lesion (OR:6.036,95 % CI:2.119-17.190,P =0.001) were the independent risk factors contributing to PMD.Conclusion Internal watershed infarction and BAD lesion may predict the PMD occurrence.The infarct location is conducive to find the high risk population of cerebral infarction progress.

ABSTRACT

OBJECTIVE:To study varieties,indications and usage features of Tibetan medicine preparations,and provide refer-ence for the clinical treatment and new drug development for Tibetan medicine. METHODS:Tibetan medicine preparations in offi-cial standards were collected,variety features(formula scale,license number,pharmaceutical enterprise,name and dosage form, herbal medicine number),action features(indication name,clinical application)and medicine features(category and quantity,me-dicinal part,commonly used drug,frequency,three fruitsuse feature) were summarized. RESULTS:Totally 458 preparations were included,including 18 recorded in Chinese Pharmacopoeia (part 1,2015 edition),200 recorded in Pharmaceutical Stan-dards of the Ministry of Health of China(Tibetan medicine,1995 edition),240 recorded in Tibetan Medicine Standards(six prov-inces). 129 had obtained 430 license numbers,from 52 pharmaceutical companies;the prearations were mainly named byherbal medicine+principal medicine+dosage formprincipal medicine+dosage formandindication+dosage form. Dosage forms had powder,pill,cream,oil,syrup and wine; the number of herbal medicine was(13.81±13.28). The Tibetan preparations were com-monly used in the clinical treatment of diseases of the digestive system,respiratory system,genitourinary system,circulatory sys-tem,infectious and parasitic diseases,etc. 442 medicines had been used,including 297 herbal medicines,70 animal medicines, 47 mineral medicines and other 28 medicines. The common medicinal parts were seed or fruit,root or rhizome,whole grass,flow-er,stem,over-ground part,tuber or bulb,etc. The top 10 frequency of medicines were as follows as Terminalia chebula,Cartha-mus tinctorius,Aucklandia lappa,Amomum kravanh,Phyllanthus emblica,Piper longum,Inula racemosa,Punica granatum, Moschus berezovskii,Adhatoda vasica;and 19% preparations contained three fruits(T. chebula,P. emblica,Terminalia bellirica). CONCLUSIONS:Tibetan medicine preparations have rich varieties,reasonable herbal medicines,more solid preparations,widely clinic application,contain more herbal drugs and commonly usethree fruits,while fewer preparations have obtained license number.

ABSTRACT

BACKGROUND:Genetic modification by Shootin1 aims to effectively improve neural differentiation of bone marrow mesechymal stem cel s (BMSCs) in the injured spinal cord, thereby promoting functional recovery from spinal cord injury after cel transplantation. OBJECTIVE:To explore the nerve regeneration ability of transplanted BMSCs overexpressing Shootin1 in rats with spinal cord injury. METHODS:BMSCs were transfected using adenovirus-Shootin1 for 48 hours. Then, immunofluorescence staining was used to detect Nestin and NeuN expression levels in the transfected cel s under in vitro neuronal induction and differentiation. Animal models of spinal cord injury were made in rats using modified Al en’s method. Thirty minutes later, Shootin1-transfected BMSCs and non-transfected BMSCs were respectively injected into the subarachnoid space of the rats in the transfection and non-transfection groups, respectively. Rats in the model group were given no treatment. Five weeks after modeling, spinal cord samples were taken from each rat to make frozen sections for detection of nerve related markers RESULTS AND CONCLUSION:After 48-hour adenoviral transfection, Shootin1 expression was successful y detected in BMSCs. After 7-day in vitro induction, the cel morphology in the three groups varied, and there was no significant difference in the expression of Nestin and NeuN between the transfection and non-transfection groups. Basso, Beattie and Bresnahan scores were higher in the two cel transplantation groups than the model group. Increased expression levels of Nestin, NeuN, GFAP, MAP-2, ChAT and SYN were observed in both two cel transplantation groups, indicating a strengthened ability of nerve regeneration. Our experimental findings further confirm that BMSCs transplantation for spinal cord injury has achieved good outcomes, and Shootin1 protein plays a certain role in nerve regeneration and functional recovery after spinal cord injury. However, Shootin1 overexpression shows no obvious additional effects in combination with BMSCs transplantation, and further studies on the optimization of BMSCs transplantation for spinal cord injury are necessary.

ABSTRACT

@#Objective To design a device for the patients with amyotrophic lateral sclerosis (ALS), who are disable to speak and move, to call for the nurses independently when necessary. Methods The electroencephalographic signals were collected and processed with TGAM, and the extracted attention characteristic values were transmitted to a computer with Bluetooth. The loudspeaker would call the nurses when the attention characteristic value exceeds the normal range. Results In the testing process of 20 participants in 10 tests, the success rate was 81.5%, and the average misjudged frequence was 0.2 within 20 min. Conclusion The device can be used in the nursing of the patients with ALS to meet the needs of daily nursing work.

ABSTRACT

BACKGROUND: Previous studies have demonstrated that acidic fibroblast growth factor (aFGF) combined with partially deproteinized bone (PDPB) (aFGF/PDPB) well promotes avascularization in animals with early-stage avascular necrosis of the femoral head (ANFH), but the histological results remain unknown.OBJECTIVE: To observe the histological repairing effects of aFGF/PDPB on early-stage ANFH in rabbits. METHODS: New Zealand rabbits were established models of bilateral ANFH and were randomly divided into a blank group, a simple PDPB group, and an aFGF/PDPB group. PDPB and aFGF/PDPB bone were implanted into the PDPB and aFGF/PDPB group accordingly. The blank group did not receive any implantation. At 2, 4, and 8 weeks after surgery, all animals were sacrificed for histological examination to observe the osteogenesis by hematoxylin-eosin staining.RESULTS AND CONCLUSION: Defects were filled with granulation tissues and fibrous connective tissues, only a little osteoid tissue formed at the borderline in the blank group at the end of the 8th week. In the PDPB group, a little new bone and cavitas medullaris formed. At 8 weeks, lots of graft was absorbed and cavitas medullaris formed with more osteoplasts and myeloid cells in it. The osteogenesis in the aFGF/PDPB group was better than that of PDPB group in each time point. At 4 weeks, the transplanted cavity was filled with osteoid tissues, a lot of osteogenic precursor cells and osteoblasts could be seen. Plenty of micrangium was observed, and osteoid tissues began to rebuild. At 8 weeks, the graft was replaced by bone tissues, and cavitas medullaris were formed with lots of bone marrow cells in it. At the borderline of the bone trabecula, there were lots of osteoplast and little osteoclasts, which may play a role in bone remodeling. There were mature bone cells in bone lacuna. Results indicate that aFGF/PDPB has better repair effect on rabbit model of ANFH than that of simple PDPB.

<p><b>OBJECTIVE</b>To find out the optimal nitrogen application level of Desmodium styracifolium.</p><p><b>METHOD</b>A field experiment using randomized block design was carried out to study the effects of 5 nitrogen application levels (150, 187.5, 225.0, 262.5 and 300.0 kg x hm(-2)) on yield and active component content of D. styracifolium.</p><p><b>RESULT</b>Nitrogen application could increase the yield and contents of polysaccharide, total flavonoides and total saponins of D. styracifolium. However, the enhancing extent of the active component content and the yield were not always significant with the increase of nitrogen level. In which, the yield were not significantly different among the nitrogen application levels of 225.0, 262.5, 300.0 kg x hm(-2) the polysaccharide content was no significantly difference among the nitrogen application levels of 225.0, 262. 5 and 300.0 kg x hm(-2), the total flavonoides content under the nitrogen level of 300.0 kg x hm(-2) was significantly lower than that of 150.0 kg hm(-2) (P < 0.01), and the total saponins content under the nitrogen level of 300.0 kg x hm(-2) was no significant difference compared with that of 262.5 kg x hm(-2).</p><p><b>CONCLUSION</b>The optimal nitrogen application level of D. styracifolium was 225.0-262.5 kg x hm(-2).</p>

ABSTRACT

BACKGROUND:Acidic fibroblast growth factor (aFGF) possesses good effects on vascularization and osteogenesis.But whether aFGF can promote the vascularization in animals with early-stage avascular necrosis of the femoral head (ANFH) remains unclear.OBJECTIVE:To investigate the vascularization of aFGF composited by partially depreteinized bone (PDPB) in repair of early-stage ANFH.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed in the College of Life Science,Nanhua University between January 2008 and January 2009.MATERIALS:Ribs from healthy,adult,New Zealand rabbits were prepared into PDPB by a series of physico-chernical methods including degreasing,deproteinization,partial decalcification and freeze drying,aFGF diluted with sterile distilled water was composited by PDPB particles to prepare artificial composite bone.METHODS:A bone window was made at the juncture of femoral head and femoral neck bilaterally in 24 healthy,adult,New Zealand rabbits.Rabbit models of bilateral ANFH were established by removing approximately 50% of cancellous bone and perfusion with 95% ethanol.Successful bilateral ANFH models were randomly divided into a blank group,a simple PDPB group,and an artificial composite bone group.PDPB and artificial composite bone were implanted into the PDPB and artificial composite bone group accordingly.The blank group did not receive any implantation.MAIN OUTCOME MEASURES:At 2,4,and 8 weeks after surgery,ink-injected specimens were prepared for microvessel count and microvessel area analysis.RESULTS:Microvessel number and microvessel area were least in the blank group,followed by simple PDPB group,and lastly the artificial composite bone group.There was significant difference in microvessel number and micrevessel area between artificial composite bone group and blank,simple PDPB groups (P<0.05).CONCLUSION:Tissue-engineered artificial bone composited by aFGF and PDPB promotes vasculadzation in repair of earlv-staqe ANFH in rabbits.

ABSTRACT

Objective To study the nursing of bedsore prevention in patients with unstable pelvic fracture. Methods Bedsore forewarning risk assessment went on in 39 patients with unstable pelvic fracture by Braden scoring method. Results No bedsore was observed in these 39 patients with unstable pelvic fracture.The nursing result turned out to be satisfactory. Conclusions The incidence rate of bed-sore can be degraded in patients with unstable pelvic fracture by forewarning risk assessment with Braden scoring method and early nursing intervention.

ABSTRACT

Objective To study influence of nursing intervention on pain and satisfaction degree of pa-tients after lower limb fracture. Methods 200 patients of lower limb fracture were divided into the experi-mental group and the control group randomly with IOO patients in each group. Routine nursing was given to the contrtol group and special nursing intervention besides routine nursing to the experimental group. Pain and sat-isfaction degree was compared between the two groups and the results underwent χ2 test. Results The pain degree in the experimental group was much less than that of the control group, the satisfaction degree of the ex-perimental group was much higher than that of the control group. Conclusions Nursing intervention can re-liege pain degree of patients with lower limb fracture,make them go through postoperation pain stage and in-crease satisfaction degree with nursing.

ABSTRACT

Objective To investigate the effect of rosiglitazone on the expression of platelet CD40 ligand (CD40L) in insulin-resistant rats, and to further determine the relationship between CD40L and insulin resistance. Methods 60 healthy male SD rats [(200±20)g] were randomly divided into 4 groups: control group (C), high fat group (HF), low dose rosiglitazone group (LR) and high dose rosiglitazone group (HR). Rats in group C were fed normal chow diet, and the others were given high fat chow diet. After 12 weeks, high dose of rosiglitazone (10mg/kg) was given to rats in group HR and low dose of rosiglitazone (5 mg/kg) was given to rats in group LR for 4 weeks. Rats in group HF and group C were given 0.9% sodium chloride solution. The level of sCD40L was measured by ELISA and the expression of platelet membrane CD40L was detected by immunoprecipitation and Western blot. The insulin resistance (IR) index was calculated by homeostasis model assessment (HOMA). Results HOMA-IR, sCD40L level and platelet membrane CD40L expression were higer in group HF than in group C (9.8±3.2 vs. 5.9±1.7, 367.3 ±35.3 vs. 232.3±120.6, 2.1±0.4 vs. 1.4±0.2, respectively, all P<0.05). Compared with the group HF, HOMA-IR, sCD40L level and platelet membrane CD40L expression were obviously decreased in group HR(5.4±1.1, 276.9±54.0, 1.4±0.3, respectively, all P<0.05), and there were no significant differences in HOMA-IR, sCD40L level and platelet membrane CD40L expression between group HF and group LR (P>0.05). Conclusions In insulin-resistant rats, the level of sCD40L and the expression of platelet membrane CD40L were higher. After treatment with high dose of rosiglitazone, sCD40L level and platelet membrane CD40L expression were decreased with the improvement of insulin resistance.

ABSTRACT

This study sought to find out a good way for the cryopreservation of tendon seeding cells so as to facilitate the preparation of tissue engineering tendons as products. The related questions are how different factors affect cell survival rate at the procedure of preservation and whether cryopreservation affects seeding cells' biological characters as well as collagen secretive function. The results of experiment indicate that DMSO is a more effective cryoprotectant in cryopreservation of tissue engineered tendon seeding cells. Blood serum nourishment is very important in cell culture, preservation and treatment. The same sustenance after cryopreservation increases cell survival rate. In the process of cryopreservation, the concentration of cells is important to cell survival rate; cell survival rate will decrease when it is less than 1.0 x 10(6)/ml. In the process of cryopreservation, the cooling speed is also important to cell survival rate, slow cooling method achieves higher cell survival rate than does the rapid cooling method. Cryopreservation by use of 10%DMSO+15%FCS+75%DMEM does not affect seeding cells' collagen secretive function greatly and does not affect seeding cells' growth curve, cell cycle and chromosome mode obviously. The prescription of 10%DMSO +15%FCS+75%DMEM is suited for the cryopreservation of tendon seeding cells.

ABSTRACT

Cell adhesion is a basic and very important tissue in the field of tissue engineering. Fibronectin and integrins are the most important elements to cell adhesion. Some surface receptors of fibroblast can also conjugate with type I collagen in extracellular matrix (ECM) directly. Laminin receptors on the surface of fibroblast bound to laminin also play a role in cell adhesion. In this paper are reviewed a number of related articles. The structures and function of fibronectin and integrins are discussed in detail; the tendon cell's adhesion structures are also discussed. Yet, there was scarcely any paper on the effects which the preservation of tissue engineered products may have on cells' adhesion fo ECM. Therefore, researching on cell adhesion and finding a way of preservation that has no or very little adverse effect on cell adhesion is an important topic. Results from expected advanced researches on cell adhesion may probably find promising applications in the field of tissue engineering.
